Nanog also known as Homeobox transcription factor Nanog is a homeobox transcription factor belonging to the Nanog homeobox family with a homeobox DNA-binding domain. Nanog is known to play a crucial role in the second embryonic cell fate specification following formation of the blastocyst. It is expressed in the inner cells of morula, blastocyst, early germ cells, embryonic stem cells (ES cells) and embryonic germ cells and expression decreases with ES differentiation. Ablation of Nanog resulted in failure in specification of early embryo pluripotent cells. Studies on ES cells have revealed that overexpression of Nanog eliminates the necessity of LIF (Leukocyte inhibiting factor) or STAT3 activation for maintaining the pluripotency. Overexpression of Nanog also prevented the ES cells to differentiate, whereas removal of Nanog directed the cells to differentiate into parietal/visceral endoderm. Studies have revealed that Nanog and STAT3 work independently whereas; Nanog and Oct4 work in coherence for maintaining ES identity and potency. It has been shown that the tumour suppressor p53 binds to the promoter of NANOG and suppresses its expression after DNA damage in mouse embryonic stem cells. Nanog has been found to be dispensable for generation of induced pluripotent stem cells, though it remains to be one of the critical transcription factors in maintaining the pluripotency of ES cells. Blocks bone morphogenetic protein-induced mesoderm differentiation of ES cells by physically interacting with SMAD1 and interfering with the recruitment of co activators to the active SMAD transcriptional complexes. Nanog a nuclear protein is generally expressed in testicular carcinoma and derived germ cell tumors and also in ovary teratocarcinoma cell line and testicular embryonic carcinoma. However expression is not common in many somatic organs and oocytes.
